Verdict is in. It is the rear wheel. I swap the campy wheel in then all is quiet.

Question is what I can do to nail down the source. Spokes seem right. No noise while cranking on stand. It is a 45mm carbon wheel so spokes are inside the rim and under rim tape. Do I need a special spoke wrench to service it?

I don't think this is spoke related, because it is very consistent and correlates to pedal stroke, i.e. chain tension. I would take out the cassette, clean, lubricate and re-install with proper torque on the lockring.

If that doesn't solve it, look into wheel and freehub bearings, or perhaps pawls.

Could even be a cracked cog, or cracked last cog/lock ring.

Are the requisite spacers for cassette/freehub body correctly in place? disregard if Campy freehub....

I will add that I also had this happen when my lock ring on the cassette had come a little loose.

Great work finding the cause...noises like that are hard to pin down sometimes.
I used to have a Ti Litespeed that developed a tick...drive me nuts. I eventually had LBS look at it because I couldn’t figure it out...after tearing most of the bike apart and test riding 3 or 4 times...they found the problem. Headset needed ti prep and be reinstalled.

My money is on the QR skewer. Some skewers get stuff really tight and some not-so-much. I'd bet that the one in the noisy wheel is the 'not-so-much' type and that there's movement of the axle in the drops.

It might be worth keeping the old wheel in but swapping just the skewer.

Thanks. My first impulse is to take out cassette, clean, inspect freehub body, then torque the lock ring to spec.

Hope that solves the problem.

In the end I think Dave's bet is correct. It is very likely the skewers.

I clean and grease the hub and reinstalled cassette. Noise is less but still there.

I tightened the skewers a bit more and less noise. So I tightened to as tight as I could the noise is gone. There is an occasional squeak here and there but it is mostly quiet now. I am surprised that the same skewers plus record hub require much less clamping force than the CK hubs.

And thinking back this probably started when I changed my skewers from campy to Enve Ti. I also had the rattling valve stem extender at that time so it might have diverted my thoughts.

I did swap the skewers back and forth but I don't recall using as much force to clamp down.

Thank you all for the suggestions. As I mentioned before this community is so helpful and helped the learning process.

Before everyone goes nuts here - if it's the wheel, I've had this with certain wheel sets and it's the first thing I check - usually to remove the freehub you need 2 allen wrenches (usually 5 mm). Many times one of the end caps is loose. Once it's tightened, the noise goes away. One time I had to remove, then reinstall the freehub to get it to stop.
