Yes, but do not cross big-big. 53-26 works. 53-29 Kaboom.

Knowledge gathered from this board. I did not get in the 53-29 yet, and I wish I won't becase there is not enough chain for that, and the rear der is too close to the 29; something *will* break if I try.
Basically I cut the chain the std length, not considering the 29. Works a treat.

The same experience. Spring for the medium cage derailleur.
Works well in either 9 spd or 10 spd. Currently running a cobbled up 11-29 cassette (from loose Veloce cogs) on my 9 spd compact crank set-up---with the medium cage---and it works beautifully. Standard chain length, as you'd use without that big 29 cog.
Actually, it will work better on some frames than others. The perfect chainstay length can gain up to 2T of wrap compared to the worst length.
It's smarter to use a medium or long cage RD.
If you do try it, set the chain length to the longest possible in the 39/13 combo. The chain should not hang loose of rub on the upper pulley chain guide in this combo.
If you never shift into the 53/29, the RD won't care if there's an extra cog.

I've been running campy record with a 13-29 cassete and a short cage rear derailleur for 5 years, and I find myself using the 53-29 gear frequently. In fact, its my favorite gear--my derailleur happens to be silky smooth in that gear. And my chain life has has not been adversly affected. I just replaced a chorus chain after 4,000+ miles--even though it showed no signs of stretch(as measured with a Park chain stretch tool).

I think Dave is on the money. The 29 + small cage works on my Serotta HSG but not at all on my Bianchi 928. A medium would be a much better fit.
[edit] Another variable I have, the derailleur on the HSG is Centaur and on the Bianchi is Chorus. Maybe that is a factor
