If i get a Shimano compatable 11 speed wheel, would a 10 speed cassette work on it with a spacer?

Yes, a lot of hubs/wheels will come with the spacer too.

I just wanted to make sure as some are listed 11speed, some 10/11 speed when listed.
I guess even without spacers in the box, I can get them.

make sure you get the right spacers as the one for Sram is a bit narrower (less thick) than the one for Shimano

I believe you will need 2.85 mm of spacers.
